    Este documento es una de las consecuencias del proyecto de cooperación entre Puertos del Estado y el Departament de Matemàtica aplicada III de la Universitat Politècnica de Catalunya (UPC) para el desarrollo de una metodología de verificación de Nivel III basada en el método de Monte Carlo. La motivación principal del estudio es la necesidad de estimación de la vulnerabilidad de un dique vertical, como paso fundamental en la metodología de verificación. La Vulnerabilidad de un dique vertical se puede definir como la probabilidad de que se produzca un determinado nivel de daño o desenlace dado que se ha producido una determinada acción. El nivel de daño es descrito por el fallo global, y la acción por un descriptor global. El descriptor será la altura de ola significante máxima. A partir de este descriptor y de otros parámetros complementarios, el método generará borrascas caracterizadas por una descripción de cada ola individual por medio de ocho parámetros no independientes entre sí. El fallo global se determinará a partir de los nueve modos de fallo considerados.     Load-independent characterization of trade-off fronts for operational amplifiers

    Abstract—In emerging design methodologies for analog integrated circuits, the use of performance trade-off fronts, also known as Pareto fronts, is a keystone to overcome the limitations of the traditional top-down methodologies. However, most techniques reported so far to generate the front neglect the effect of the surrounding circuitry (such as the output load impedance) on the Pareto-front, thereby making it only valid for the context where the front was generated. This strongly limits its use in hierarchical analog synthesis because of the heavy dependence of key performances on the surrounding circuitry, but, more importantly, because this circuitry remains unknown until the synthesis process. We will address this problem by proposing a new technique to generate the trade-off fronts that is independent of the load that the circuit has to drive. This idea is exploited for a commonly used circuit, the operational amplifier, and experimental results show that this is a promising approach to solve the issue

    Protein kinase A activity is required for the budding of constitutive transport vesicles from the trans-Golgi network

    We have examined the role played by protein kinase A (PKA) in vesicle-mediated protein transport from the trans-Golgi network (TGN) to the cell surface. In vivo this transport step was inhibited by inhibitors of PKA catalytic subunits (C-PKA) such as the compound known as H89 and a myristoylated form of the inhibitory peptide sequence contained in the thermostable PKA inhibitor. Inhibition by H89 occurred at an early stage during the transfer of vesicular stomatitis virus G glycoprotein from the TGN to the cell surface. Reversal from this inhibition correlated with a transient increase in the number of free coated vesicles in the Golgi area. Vesicle budding from the TGN was studied in vitro using vesicular stomatitis virus-infected, permeabilized cells. Addition to this assay of C-PKA stimulated vesicle release while it was suppressed by PKA inhibitory peptide, H89, and antibody against C-PKA. Furthermore, vesicle release was decreased when PKA-depleted cytosol was used and restored by addition of C-PKA. These results indicate a regulatory role for PKA activity in the production of constitutive transport vesicles from the TGN

    How to Improve Sports Fans' Attitudes Toward the Sponsor Through Brand Management? A PLS and QCA Approach

    The aim of the article is to explain attitudes towards the sponsors of a sporting event from brand management, especially considering the perceptions of congruence with the sponsor, quality, value, and two less common variables of innovation and popularity. The analysis has been carried out using two methodological approaches: a Partial Least Squares (PLS) model and a Qualitative Comparative Analysis (QCA). PLS results indicate that congruence, innovation and popularity significantly predict attitudes towards the sponsor, explaining up to 61% of it. On the other hand, QCA analysis shows nine interactions capable of producing the expected result, where congruence, quality innovation and popularity have shown a relevant role. This study has implications at a theoretical and practical level, contributing to understanding consumer behaviour in the context of sporting events and providing marketing managers with valuable information to help improve the performance of their sponsorships.Ciencias de la Actividad Física y del Deport

    Trimeric G proteins modulate the dynamic interaction of PKAII with the Golgi complex

    The Golgi complex represents a major subcellular location of protein kinase A (PKA) concentration in mammalian cells where it has been previously shown to be involved in vesicle-mediated protein transport processes. We have studied the factors that influence the interaction of PKA typeII subunits with the Golgi complex. In addition to the cytosol, both the catalytic (Cα) and regulatory (RIIα) subunits of PKAII were detected at both sides of the Golgi stack, particularly in elements of the cis- and trans-Golgi networks. PKAII subunits, in contrast, were practically absent from the middle Golgi cisternae. Cell treatment with either brefeldin A, AlF4 − or at low temperature induced PKAII dissociation from the Golgi complex and redistribution to the cytosol. This suggested the existence of a cycle of association/dissociation of PKAII holoenzyme to the Golgi. The interaction of purified RIIα with Golgi membranes was studied in vitro and found not to be affected by brefeldin A while it was sensitive to modulators of heterotrimeric G proteins such as AlF4 −, GTPγS, βγ subunits and mastoparan. RIIα binding was stimulated by recombinant, myristoylated Gαi3 subunit and inhibited by cAMP. Pretreatment of Golgi membranes with bacterial toxins known to catalyze ADP-ribosylation of selected Gα subunits also modified RIIα binding. Taken together the data support a regulatory role for Golgi-associated Gα proteins in PKAII recruitment from the cytosol

    Video Quality Assessment needs to correspond to human perception. Pixel-based metrics (PSNR or MSE) fail in many circumstances for not taking into account the spatio-temporal property of human's visual perception. In this paper we propose a new pixel-weighted method to improve video quality metrics for artifacts evaluation. The method applies a psychovisual model based on motion, level of detail, pixel location and the appearance of human faces, which approximate the quality to the human eye's response. Subjective tests were developed to adjust the psychovisual model for demonstrating the noticeable improvement of an algorithm when weighting the pixels according to the factors analyzed instead of treating them equally. The analysis developed demonstrates the necessity of models adapted to the specific visualization of contents and the model presents an advance in quality to be applied over sequences when a determined artifact is analyzed

    Insertion of impairments in test video sequences for quality assessment based on psychovisual characteristics

    Assessing video quality is a complex task. While most pixel-based metrics do not present enough correlation between objective and subjective results, algorithms need to correspond to human perception when analyzing quality in a video sequence. For analyzing the perceived quality derived from concrete video artifacts in determined region of interest we present a novel methodology for generating test sequences which allow the analysis of impact of each individual distortion. Through results obtained after subjective assessment it is possible to create psychovisual models based on weighting pixels belonging to different regions of interest distributed by color, position, motion or content. Interesting results are obtained in subjective assessment which demonstrates the necessity of new metrics adapted to human visual system
